Morning Session: HPLC Basics & Troubleshooting
• HPLC Components & Equipment : Mobile phase, Pumps, Autosamplers, Detectors
• HPLC terms: Retention time, retention factor, separation factor, resolution
• Troubleshooting:
- Common problems
- Baseline drifting, noisy, spiky, cycling etc.
- Back Pressure – high, low, fluctuating
- Void Volume peak – retention time, size, shape
- Peak Retention times – early, late, variable
- Peak Height and Shape – normal, tailing, shoulders, split peaks
Afternoon Session: Column Chemistries & Method Development
• HPLC Column Packing Material (Solid support, Stationary phases)
• Column Chemistries
• Separation mode with a focus on reversed-phase
• Separation challenges
• Pellicular packing
The first part of this course will guide you through the different components of an HPLC system and related troubleshooting issues and solutions.
The second part of the day will be detailing the different types of column chemistries with their method development applications.
This course will be especially valuable to HPLC users with less than 5 years’ experience or anyone who has not done HPLC for a while and wants a refresher.
Themis Flarakos is a chromatography expert with over 20 years’ experience and has developed hundreds of methods. During these sessions, Themis will be sharing his experience and answering all of your questions.
